Figure 7.A.3. Dideoxy sequencing. Step 3: Add replication stuff to start synthesis of the complementary strand of DNA. The primary ingredient here is a polymerase enzyme. The polymerase grabs free nucleotides, reads the next next nucleotide on the single-stranded part of the DNA, and then glues the appropriate base pair on the complementary strand. For example, if the next nucleotide on the black strand is T, then the polymerase will glue an A onto the end of the brown strand.
